Phenotypic Signature of Osteosarcoma

Jeanette Loutsch, PhD

Arkansas State University

Abstract

Osteosarcoma (OS) is a rare tumor of the bone and soft tissue found usually in adolescent children and even rarer in adults. This tumor has a poor prognosis when metastasis is present at time of diagnosis. It is the goal of this research proposal to investigate the phenotypic signature of OS in adult and adolescent onset. This will be accomplished by performing cDNA microarray analysis of tumors at the UAMS tissue bank with validation of the genes that were either expressed in at a higher or lower level than normal bone tissue. The validation of the gene product will consist of immunohistochemical analysis of tumor sections and cultured OS cells, Saos2 with additional RT-PCR of the genes expressed in the Saos2 cell line. These preliminary studies will aid in the understanding of the tumor progression.
