Evaluation of Thiazolidines for Ability to Prevent Oxidative Damage to Membrane Lipids

Abstract

Five thiazolidines which we have synthesized in our laboratory will be evaluated for ability to prevent oxidative damage to membrane lipids using three models. Liposomes will be prepared to simulate membrane lipids. In the first and second models, liposomes will be exposed to hydroxyl radical generated from 2,2-azobis-(amidinopropane) hydrochloride (AAPH) and will be assayed for oxidative damage using the TBARS  and N-methyl-2-phenylindole tests. In the third model, liposomes will be exposed to molecular oxygen and oxygen uptake and conjugated diene formation will be monitored. This model allows kinetic parameters to be calculated.
